The Macroautophagy Machinery in MHC Restricted Antigen Presentation

نویسندگان

چکیده

Autophagy-related (ATG) gene products regulate macroautophagy, LC3-associated phagocytosis (LAP) and LC3-dependent extracellular vesicle loading secretion (LDELS). These processes also influence antigen processing for presentation on major histocompatibility complex (MHC) molecules to T cells. Here, I summarize how these different pathways use the macroautophagy machinery, contribute MHC class II restricted autoimmunity, tumor immunology immune control of infectious diseases. Targeting should allow regulation intracellular cells modulate protective pathological responses.

Antigen Processing by Macroautophagy for MHC Presentation

T cells recognize antigen fragments, presented to them by MHC molecules. It lies in the interest of the immune system to display a maximal diversity of these peptides and utilize all catabolic processes to generate them. Macroautophagy, a pathway that delivers cytoplasmic constituents for lysosomal degradation is no exception. In recent years, it has become apparent that macroautophagy assists ...

Metformin Suppresses MHC-Restricted Antigen Presentation by Inhibiting Co-Stimulatory Factors and MHC Molecules in APCs

Metformin is widely used for T2D therapy but its cellular mechanism of action is undefined. Recent studies on the mechanism of metformin in T2D have demonstrated involvement of the immune system. Current immunotherapies focus on the potential of immunomodulatory strategies for the treatment of T2D. In this study, we examined the effects of metformin on the antigen-presenting function of antigen...

Macropinocytosis in phagocytes: regulation of MHC class-II-restricted antigen presentation in dendritic cells

Dendritic cells (DCs) are outstanding antigen presenting cells (APCs) due to their robust ability to internalize extracellular antigens using endocytic processes such as receptor-mediated endocytosis, phagocytosis, and macropinocytosis. Macropinocytosis mediates the non-specific uptake of soluble antigens and occurs in DCs constitutively. Macropinocytosis plays a key role in DC-mediated antigen...

Mechanisms of interference with the MHC class I-restricted pathway of antigen presentation by herpesviruses.

Herpesviruses are an ancient, ubiquitous family of DNA viruses, most of which share a lifestyle of latently or persistently infecting a young host, and spreading to infect a new host a generation later. Most herpesviruses interfere with antigen presentation via the MHC class I-restricted pathway of antigen presentation, suggesting that impairment of the cytotoxic T lymphocyte response is necess...

Efficient and qualitatively distinct MHC class I-restricted presentation of antigen targeted to the endoplasmic reticulum.

For most nascent glycoprotein Ags, the MHC class I-restricted processing pathway begins in the endoplasmic reticulum (ER). From this location, they are translocated to the cytosol for degradation by the proteasome. A reasonable assumption is that processing of exocytic Ags is less efficient than that of cytosolic Ags, due to the requirement for additional handling, but that the processing pathw...
